Synthetic melanocortin receptor agonist researched for sexual dysfunction, arousal, and libido enhancement in both male and female subjects.

PT-141 (Bremelanotide) is a synthetic peptide derived from Melanotan-2 that acts as a melanocortin receptor agonist, primarily targeting the MC3R and MC4R receptors in the central nervous system. Unlike PDE5 inhibitors that work on vascular mechanisms, PT-141 acts directly on the brain’s arousal pathways to influence sexual desire and response.
Originally developed during Melanotan-2 research when subjects reported significant increases in sexual arousal, PT-141 was isolated and refined specifically for its effects on sexual function. It received FDA approval in 2019 as Vyleesi® for hypoactive sexual desire disorder (HSDD) in premenopausal women, making it the first melanocortin-based therapeutic approved for sexual dysfunction.
PT-141 crosses the blood-brain barrier and binds to melanocortin-4 receptors (MC4R) in the hypothalamus and limbic system. This activation triggers downstream signaling cascades that modulate dopaminergic and oxytocinergic pathways involved in sexual arousal and desire.
MC4R activation by PT-141 increases dopamine release in the medial preoptic area (MPOA) and nucleus accumbens, brain regions directly associated with sexual motivation and reward. This mechanism explains why PT-141 influences desire itself rather than just physiological response.
Research indicates PT-141 stimulates oxytocin release from the paraventricular nucleus of the hypothalamus. Oxytocin plays a role in sexual arousal, bonding, and the physiological reflexes associated with sexual response in both sexes.
Because PT-141 works through central melanocortin pathways rather than peripheral vascular mechanisms, it demonstrates efficacy in both male and female research subjects – a distinction from vascular-targeted compounds that primarily benefit male subjects.
